Route 53 provides local service throughout Upper Marlboro, Marlboro Village, and Marlboro Meadows. Key stops include the County Department of Corrections, Marlboro Village Shopping Center, the County Courthouse, the County Administrative Building, and residences along Village Drive West.

Effective December 4, 2023, Route 53 no longer stops in the Equestrian Center Park and Ride. It also has major timetable adjustments.
